cDNA Cloning of the Genes Induced in thd Ischemic Human Liver

Abstract
A cDNA library was made from polya+ RNA of human liver extracted 30 minutes after ischemic injury. Differential screening of the cDNA library with the two first-strand cDNA probe from normal liver and from ischemic liver, resulted in isolation of

ischemia-induced cDA clones named as pHSL 41 and pHSL 42. Cross hybridization between the two clones revealed that they may represent 2 different genes. Expression analysis of the genes demonstrate that they are considerably induced in the
ischmic
liver
compare to the normal liver. Partial sequence analysis of the clones demonstrated that pHSL 41 did not show any significnat homology to other known genes in the gene data base and may represent novel ischemia-induced gene in the liver. However,
the
clone pHSL 42 was proven to be a human triglyceride lipase gene.
